Hole in slick

The question is can I patch a small hole in a slick?

I run tubes and screws and was curious if it safe to removed the tire and reinstall after being screwed?

My main concern is the rim screws. It it possible to break the bead on a tire with the one side still screwed. I would like the screws to go back in the same holes on the tire.
